One Perkinsus species may hide another: characterization of Perkinsus species present in clam production areas of France ArchiMer

Although clam populations in France are known to be infected with protozoans of the genus Perkinsus, no molecular characterization was previously performed on these parasites. Considering that several members of this genus have been associated with mortalities of molluscs worldwide, a study was undertaken in order to characterize these parasites in France. For that purpose, clams, Ruditapes philippinarum and R. decussatus, collected from different production areas and found to be infected with Perkinsus sp. in thioglycolate culture medium, were selected for PCR-RFLP tests and sequencing. Optimisation de l'élevage intensif de la palourde japonaise Ruditapes philippinarum en marais ArchiMer

Après les études de mise au point de la technique intensive réalisées en 1988 et 1989 sur des bassins expérimentaux de 30 m2, une optimisation de cette méthode d'élevage a été faite sur deux claires de 200 m2 avec les normes zootechniques suivantes: densité = 300 palourdes x m-2, concentration d'eau de mer enrichie en Skeletonema costatum produit à partir d'eau salée souterraine = 300 cellules/µl ou 126 µg/l de chla, taux de renouvellement = 100 % du volume par jour. Perspectives de développement de l'élevage de la palourde japonaise Ruditapes philippinarum dans le bassin ostréicole de Marennes-Oléron ArchiMer

The complete Manila clam's breeding cycle (Ruditapes philippinarum ) in oysters ponds (claires) permits to obtain a length of 25 mm after 1 year with a density range of 200 animals/m2 . As the primary production is restricted, the breeding biomass during the second year is limited to 500 g.m2 corresponding to a density between 30 to 50 clams..m-2. For an increase of the production it is necessary to extend the cultivated area to the tida1 bottom of the Bay of Marennes-Oleron. It appears that the man tidal level between + 2 and + 2.5 15 necessary with a density range of 200.m-2 to reach, the market size nine months later. The harvest sould be done before winter time to avoid a large 1055 of the flesh which can cause high levels of mortality.

Polyploidy in the Manila clam Ruditapes philippinarum. II. Chemical induction of tetraploid embryos ArchiMer

Tetraploidy was induced in embryos of Ruditapes philippinarum by treating the eggs with cytochalasin B. (CB). Two treatment times were found to be effective in inducing tetraplidy : 0 to 10 minutes after insemination (before formation of the first polar body) and 45 minutes after insemination (just before the first cleavage). Treatments, started 5 and 45 minutes after insemination, produced 64.4 ± 7.8% and 28.3 ± 2.3% tetraploid embrios, respectively, after 6 hours (on the average over 3 experiments). However, no tetraploids were detected in 4-month-old spat. Sperm entry delayed by the CB could be involved in induction of tetraploidy by early treatment.

Présence du bivalve Veneridae Ruditapes philippinarum (Adams & Reeve, 1850) sur le littoral sud de la Tunisie OceanDocs

Dans cette étude nous signalons la présence d’une espèce d’origine indo-pacifique, Ruditapes philippinarum, sur les côtes tunisiennes, dans le golfe de Gabès. Les échantillons prélevés ont montré des pourcentages de 89 pour l’espèce indigène Ruditapes decussatus et 11 pour l’espèce exotique Rudiapes philippinarum à Gargour (au sud de Sfax); alors que ces pourcentages sont respectivement de 21,5 et de 78,5 au nord de cette région, à El Aouabed.

The different epizooties affecting traditional or recent shellfish culture activities (hatcheries, cockle culture) brought to light some microorganisms and parasites considered as the responsible pathogenic agents. However, in a number of cases, the etiology of these massive mortalities remains unexplained (Grassostrea gigas, Ruditapes philippinarum). Some additional works, based on some electron microscopy techniques and isolation methods, are thus essential in order to precise the infectious nature of these diseases.

Reduction of adhesion properties of Ruditapes philippinarum hemocytes exposed to Vibrio tapetis ArchiMer

Vibrio tapetis is the causative agent of brown ring disease (BRD), which affects a species of clam, Ruditapes philippinarum. After incubation with V. tapetis, hemocytes lose filopods and become rounded, indicating cytotoxic activity of the bacterium. To rapidly quantify this cytotoxicity, a flow-cytometry test was developed based on the capacity of V tapetis to inhibit adhesion of clam hemocytes to plastic. Several bacteria:hemocyte ratios, the cytotoxicity of other Vibrio spp. pathogenic to bivalves, and that of various V. tapetis isolates were tested. Inhibition of adherence is detectable with as few as 5 bacteria per hemocyte. Methods of gynogenesis, androgenesis and polyploidy induction were developed with the aim of genetic improvement in the rainbow trout, the Pacific oyster, the Manila clam and the black scallop. In the trout: · haploid androgenesis was induced by means of a pressure shock applied on diploid eggs, probably at the time of karyogamy. Spatio-temporal patterns of perkinsosis in the Manila clam Ruditapes philippinarum from Arcachon Bay (SW France) ArchiMer

Pathogens belonging to the genus Perkinsus infect many bivalve molluscan species around the world, including the Manila clam Ruditapes phihppinarum. We investigated the spatial distribution of this parasite at 34 stations throughout Arcachon Bay (SW France). Prevalence of perkinsosis was 93% and mean infection abundance was 96 x 10(3) cells g(-1) wet gill. Lowest mean abundances were found close to the Leyre River mouth and a significant negative correlation was observed between mean abundance and salinity. Stable isotopes changes in the adductor muscle of diseased bivalve Ruditapes philippinarum ArchiMer

In this article, we show how a disease could bias stable isotope analyzes of trophic networks and propose a strategy in the choice of tissues to be analyzed. In the past few years, a new pathology (brown muscle disease or BMD) affecting the posterior adductor muscle of Ruditapes philippinarum has emerged in Arcachon Bay. BMD induces a necrosis of muscle tissues which become infused by conchiolin and hence calcified. The Manila clam population in Arcachon Bay (SW France): Can it be kept sustainable? ArchiMer

The venerid clam Ruditapes philippinarum is the most prominent suspension-feeding bivalve inhabiting muddy intertidal seagrass beds in Arcachon Bay (SW France). It is exploited by fishermen, and Arcachon Bay ranks number one in France in terms of production and total biomass of this species. Previous studies revealed a decrease in the standing stock of R. philippinarum since 2003 and unbalanced length-frequency distributions with a lack of juveniles and of adults >40 mm. Vibrio tapetis is a marine bacterium causing Brown Ring Disease (BRD) in the Manila clam Ruditapes philippinarum. V. tapetis biofilm formation remains unexplored depite the fact that it might be linked to pathogenicity. Our objectives were to characterize the in vitro biofilm formation of V tapetis and evaluate the effects of culture conditions. Biofilm structure and its matrix composition were examined by confocal laser scanning microscopy and scanning electron microscopy. Virus-like particles associated with brown muscle disease in Manila clam, Ruditapes philippinarum, in Arcachon Bay (France) ArchiMer

Recently, Manila clam, Ruditapes philippinarum, populations have suffered mortalities in Arcachon Bay (SW France). Mortality was associated with extensive lesions of the posterior adductor muscle, which become progressively brown and calcified. Ultrastructural observations by transmission electron microscopy revealed tissue degradation with necrotized muscle fibres and granulocytomas. Unenveloped virus-like particles (VLPs) were detected in muscle, granulocytic, epithelial and rectal cells. VLPs were abundant in the extracellular space, in the cytoplasm (free or enclosed in vesicles) and in the nucleoplasm of granulocytes. Asari (= Manila) clam, Ruditapes philippinarum, is the second bivalve mollusc in terms of production in the world and, in many coastal areas, can beget important socio-economic issues. In Europe, this species was introduced after 1973. In Arcachon Bay, after a decade of aquaculture attempt, Asari clam rapidly constituted neo-naturalized population which is now fished.
